This is a ready-to-use bridging study protocol for transferring a validated container closure integrity test (CCIT) method from a sending site to a receiving or contract fill-finish site. It works alongside USP <1224>‘s general comparative-testing framework for analytical method transfer, adding the CCIT-specific technical content a generic method transfer protocol does not carry: receiving-instrument equipment qualification against the sending site’s characterized range, a certified defect-standard bridging run at the receiving site, fixture and restraint hardware qualification for flexible or multi-seal formats, and an explicit statement that the maximum allowable leakage limit (MALL) does not move to accommodate the new site. Replace every <<FILL: ...>> placeholder with your own specifics and route it through your normal document control, review, and approval before execution. A worked filled specimen follows.

1. Purpose
To demonstrate that <<FILL: receiving site>> can execute the validated CCIT method for <<FILL: product / container system>> with detection performance equivalent to <<FILL: sending site>>, at the same maximum allowable leakage limit, before the receiving site is relied upon for CCIT results supporting release, stability, or PPQ disposition.
2. Scope
| Dimension | Decision |
|---|---|
| CCIT method and technology transferring | <<FILL: e.g. vacuum decay, HVLD, mass extraction, headspace analysis>> |
| Container closure system(s) covered | <<FILL>> |
| Receiving instrument make and model | <<FILL>> |
| Fixturing or restraint hardware in scope | <<FILL: e.g. cryobag restraint fixture, PFS dual-seal scan fixture, or N/A for a rigid vial>> |
| Out of scope | <<FILL: e.g. a change to the MALL or acceptance criteria, which this protocol does not permit>> |
3. Responsibilities
| Role | Responsibility |
|---|---|
| Analytical / packaging SME | Authors this protocol, defines the bridging acceptance criteria, and interprets the comparison between sites |
| Sending site method owner | Supplies the validated method, the original detection-rate or POD data, and the certified defect standard lot(s) used in validation |
| Receiving site lab / packaging lead | Executes the equipment qualification and the bridging run, and owns the receiving instrument going forward |
| CDMO relationship owner, where the receiving site is a contract site | Confirms the transfer result before the site is relied upon for a commercial batch, and escalates a bridging failure per the technical agreement |
| Quality Assurance | Approves the protocol and the transfer disposition; the point at which a partial bridging result gets converted into a documented gap rather than a quiet pass |
4. Prerequisites before the bridging run
| Prerequisite | Evidence required |
|---|---|
| Receiving instrument equipment qualification (IQ/OQ) | <<FILL: ref, confirming the instrument reads the same physical quantity across the same range as the sending site's instrument>> |
| Certified defect standards available at the receiving site | <<FILL: certificate reference, same nominal hole sizes or defect set used in the original validation, or an equivalent certified set with a documented equivalence rationale>> |
| Fixturing or restraint hardware qualified at the receiving site (if applicable) | <<FILL: ref, or N/A>> |
| Receiving site personnel trained on the method | <<FILL: training record ref>> |
Do not proceed to section 5 until every row above has dated evidence; a bridging run performed against an unqualified instrument or an uncertified defect standard produces a result with nothing to anchor it.
5. Equipment qualification comparison
| Parameter | Sending site instrument | Receiving site instrument | Equivalent? |
|---|---|---|---|
| Detection range (per USP <1207.2> characterization) | <<FILL>> | <<FILL>> | <<FILL: yes/no>> |
| Chamber volume / transducer sensitivity (if applicable) | <<FILL>> | <<FILL>> | <<FILL>> |
| Cycle time / parameters | <<FILL>> | <<FILL>> | <<FILL>> |
| Calibration status | <<FILL>> | <<FILL>> | <<FILL>> |
Any row scored “No” requires a documented rationale for why the bridging run can still proceed, or a corrective action closing the gap before it does.
6. Bridging run: certified defect standards at the receiving site
Run the same certified defect standards used in the sending site’s original validation, or an equivalent certified set with a documented lot-to-lot equivalence rationale, through the receiving site’s qualified instrument and, where applicable, fixture.
| Defect location | Hole size / defect (certified) | n | Sending site detection rate (from original validation) | Receiving site detection rate | Comparable? |
|---|---|---|---|---|---|
<<FILL>> | <<FILL: at or below the MALL>> | <<FILL>> | <<FILL>> | <<FILL>> | <<FILL: yes/no>> |
<<FILL>> | <<FILL: bracketing the MALL>> | <<FILL>> | <<FILL>> | <<FILL>> | <<FILL>> |
| Negative control (integral units) | N/A | <<FILL>> | 100% correctly passed | <<FILL>> | <<FILL>> |
Where a POD95/UCL95 curve was fit during the original validation, compare the receiving site’s fitted estimate against the sending site’s rather than comparing single detection-rate points alone.
7. Acceptance criteria
| Criterion | Requirement |
|---|---|
| Receiving site detection rate at or below the MALL | Matches or exceeds the sending site’s validated performance; no defect at or below the MALL goes undetected |
| Negative controls | All correctly pass at the receiving site |
| MALL and acceptance criteria | Unchanged from the sending site’s validated method; this bridging study does not establish a new MALL |
| Fixture/restraint baseline (if applicable) | Receiving site’s no-leak baseline spread characterized and shown comparable to the sending site’s |
A bridging study does not pass on defect-detection results alone if the receiving site’s equipment qualification or fixture qualification is incomplete; both have to be closed for the transfer to be considered valid.
8. Deviation handling
Any bridging run result showing a defect at or below the MALL going undetected at the receiving site is a failed transfer, not a result to average against a passing run elsewhere in the study. Open a deviation, investigate the receiving instrument, fixture, or defect standard as the probable cause before repeating, and do not retest silently outside a documented deviation.
9. Summary and disposition

Filled specimen
Extract for a vacuum decay CCIT method transferring from a sponsor’s development site to a contract fill-finish CDMO ahead of a PPQ campaign for a lyophilized biologic vial. Company, product, and numbers are illustrative.
| Field | Entry |
|---|---|
| Method | Vacuum decay, per validated protocol CCIT-2026-04 |
| Sending site | Acme Therapeutics development facility |
| Receiving site | CMO Beta, commercial fill-finish line 2 |
| MALL | 5 micrometers equivalent diameter |
Equipment qualification comparison confirmed CMO Beta’s vacuum decay instrument, a different model from the sending site’s unit, characterized to the same detection range with an equivalent chamber design; the calibration status was current, and cycle parameters were set to match the validated method exactly rather than the receiving instrument’s factory default.
Bridging run results using certified 2 and 5 micrometer laser-drilled vials, same certificate lot shipped from the sending site:
| Defect location | Hole size | n | Sending site rate | Receiving site rate | Comparable? |
|---|---|---|---|---|---|
| Stopper seal | 2 micron | 20 | 80% | 75% | Yes, within expected variability at this sub-MALL size |
| Stopper seal | 5 micron | 20 | 100% | 100% | Yes |
| Negative control | Integral | 20 | 100% correctly passed | 100% correctly passed | Yes |
Disposition: pass. Both sites detect reliably at and above the 5 micron MALL; the lower detection rate at 2 microns, below the MALL on both instruments, is consistent with the sub-MALL capability boundary already characterized in the original validation and does not affect the acceptance basis. CMO Beta authorized to generate CCIT results supporting PPQ effective the approval date below, with no change to the MALL or acceptance criteria.
Common inspection findings this protocol prevents
- A method transfer confirmed only by running integral units at the receiving site, with no positive control challenge showing the new instrument actually detects a defect at the MALL.
- A receiving instrument assumed equivalent to the sending site’s because it shares a model number, with no documented equipment qualification comparison.
- A fixture or restraint hardware used at the receiving site with no qualification of its own no-leak baseline.
- A bridging study result used to quietly renegotiate the MALL or acceptance criteria to fit what the receiving site’s instrument happened to show.
How to adapt this protocol
- Set your document number, method, sending and receiving sites, and linked CCIT study protocol in the header.
- Complete the equipment qualification comparison in section 5 before scheduling the bridging run; do not run positive controls against an instrument whose range has not been characterized.
- Use the same certified defect standard lot, or a documented equivalent, at both sites so the comparison in section 6 is not confounded by a different defect population.
- For a flexible or multi-seal container, confirm the fixture qualification prerequisite in section 4 before treating any bridging result as valid.
- Never use this protocol to change the MALL or acceptance criteria; if the receiving site cannot meet the sending site’s validated performance, that is a failed transfer requiring investigation, not a new baseline.
