PPQ Master Summary Table for Filing

A plug-and-play master summary table for process performance qualification results, one row per CQA or CPP and one column per batch, built for direct use in an S.2.5 or P.3.5 CTD section, with the opening paragraph pattern and a filled specimen.

How to use this document

Copy the template below into your quality system. Replace every <<FILL: ...>> placeholder with your company, system, and process specifics, then route it through your own document control, review, and approval. The filled specimen shows one worked example; adapt the numbers and scope to your operation and verify each cited regulation or standard against the current source before you rely on it.

This is a generic, originally worded starting point. It contains no confidential, proprietary, or employer material, it is drafted with AI assistance and reviewed, it is not legal or compliance advice, and adopting it does not by itself create or prove compliance. See the Disclaimer and Terms.

This is a ready-to-use master summary table for process performance qualification (PPQ) results, built for direct use inside a CTD S.2.5 or P.3.5 section. One master table replaces batch-by-batch narrative paragraphs that repeat the same structure with different numbers, the single most under-used device in a validation summary. Replace every <<FILL: ...>> placeholder with your own specifics. A worked filled specimen follows. Verify each cited reference against the current source before you rely on it.

1. Purpose

This document assembles the PPQ results a filing’s S.2.5 or P.3.5 section needs into a single reviewer-ready view: the validation approach stated once, one master table spanning every PPQ batch, and one explicit conclusion sentence, rather than repeated batch-by-batch narrative.

2. Opening paragraph

State the validation approach, the batch count, and the rationale in the first paragraph. A reviewer’s first move is to check whether this is stated plainly or has to be inferred from a table.

<<FILL: e.g. "Process validation for the [drug substance/drug product] manufacturing process follows [traditional process validation / continuous process verification / a hybrid approach], with process performance qualification executed across [N] commercial-scale batches. The batch count reflects [rationale, tied to process characterization work or an applicable flexibility guidance]; the residual risk is addressed through [enhanced characterization / the Stage 3 continued process verification commitments described below].">>

3. Master summary table

One row per CQA or CPP, one column per batch, the acceptance criterion stated once rather than repeated in every cell.

4. Statistical basis of the claim

State which claim is actually being made; “passed” alone is not sufficient.

5. Deviations during PPQ

State every deviation plainly; do not omit or footnote one. If none occurred, state that explicitly rather than leaving the section silent.

6. Conclusion sentence

Write the conclusion explicitly; do not let the table imply it.

<<FILL: e.g. "PPQ results demonstrate the process consistently meets the acceptance criteria defined in Section [X] for all batches manufactured to date, and the Stage 3 monitoring plan described in Section [Y] describes the ongoing verification that will continue to build confidence in the process across additional commercial lots.">>

Filled specimen

Extract for an example generic cell therapy drug substance PPQ campaign. Company, product, and numbers are illustrative, not a real product’s data.

Opening paragraph: “Process validation for the drug substance manufacturing process follows traditional process validation, with process performance qualification executed across two commercial-scale batches. The batch count reflects the total number of qualified apheresis-matched batches manufactured at commercial scale as of this submission; the residual risk of a two-batch PPQ campaign is addressed through the enhanced process characterization program described in Section 3.2.S.2.6 and through the Stage 3 continued process verification commitments described below.”

Statistical basis: release specifications are assessed as a simple pass/fail against the fixed acceptance criterion defined in Section S.4.1; a tolerance interval was not applied given the two-batch campaign size, a limitation stated directly in the Stage 3 monitoring commitment rather than implied.

Deviations: none occurred during the PPQ campaign.

Conclusion sentence: “PPQ results demonstrate the process consistently meets the acceptance criteria defined in Section 3.2.S.4.1 for both batches manufactured to date, and the Stage 3 monitoring plan in Section 3.2.S.2.6 describes the ongoing verification that will continue to build confidence in the process across additional commercial lots.”

Common inspection findings this document prevents

  • Batch-by-batch narrative paragraphs repeating the same structure with different numbers instead of one master table a reviewer can scan.
  • A table with no stated conclusion sentence, leaving the reviewer to infer the claim.
  • “Passed” asserted with no statement of which statistical claim, capability index, tolerance interval, or simple pass/fail, is actually being made.
  • A PPQ deviation omitted or left for the reviewer to find independently in the raw data.

How to adapt this document

  1. Set the CTD section reference and underlying PPQ protocol/report in the header.
  2. Use the same CQA and CPP names and numbering the rest of Module 3 uses; do not introduce a synonym here.
  3. State the statistical method actually used in section 4; do not let “passed” stand in for it.
  4. Disclose every deviation in section 5, including a stated “none occurred” when true; never leave it blank.
  5. Write the conclusion in section 6 as an explicit sentence a reviewer can cite back, not an implied takeaway from the table.
